The regulation of Lycium barbarum (LB) on apoptosis of rat spleen induced by hydrocortisone (HYD) was studied in this paper. The results indicated LB could inhibit the apoptosis induced by HYD and the inhibition is dose dependent.

[Effects of pure and crude Lycium barbarum polysaccharides on immunopharmacology]

Effects of pure Lycium barbarum polysaccharides (LBP-X) on immunological activity were compared with crude LBP. The pure LBP-X were divided into different doses, lower doses (5-20 mg/kg.d) of pure LBP-X showed a remarkable effect on immunological enhancement. Especially, LBP-X 10 mg/kg.d had a highly significance difference compared with crude LBP on immune indices in mice. Lycium barbarum polysaccharides have the best pharmacological effect when it was the best dose.

[Inhibition the growth of human leukemia cells by Lycium barbarum polysaccharide]

The effect and the mechanism of Lycium barbarum polysaccharide (LBP-X) on inhibiting the growth of human leukemia HL-60 cells were examined. LBP-X(20, 100, 500, 1000 mg/L) could inhibit the growth of HL-60 cells in dose-dependent manner and decrease the membrane fluidity of the cell. Agarose gel electrophoresis of DNA from the cells treated with LBP-X revealed a "DNA ladder" and positive TUNEL test. The results showed that the apoptosis of HL-60 cells induced by LBP-X maybe its important mechanism on anti-tumorgenesis.

[Effect of lycium barbarum L on defending free radicals of mice caused by hypoxia]

The effect of Lycium Barbarum L. (LB) on the free radical defending system during hypoxia was tested. Fifty six mice were divided into 3 groups. Experimental mice were fed with LB and the control mice were fed with distilled water by tube feeding for 16 days. Animals were put into each enclosed bottle for testing the effect of hypoxia. The results showed that LB treatment could not prolong the survival time (P > 0.05), but increase the activities of superoxide dismutase (SOD), catalase(CAT) and total anti-oxidative capacity as compared with the control group(P < 0.05). The study indicated that LB could not increase the hypoxic tolerance, but it might have a protective effect on free radical injury caused by hypoxia.

[Study on the composition of Lycium barbarum polysaccharides and its effects on the growth of weanling mice]

In order to observe the effects of lycium barbarum polysaccharides (LBP-4) on the growth of weanling mice and the absorption of some metals in the their body, the composition of LBP-4 is determined. 120 female weanling mice are divided in random into 4 groups. They are fed on LBP-4 at the dose of 5, 10 and 20 mg/(kg.d) respectively. The taken feed weight and the body weight of mice are recorded everyday. After 21 days, the content of calcium, magnesium, zinc and iron in pygal muscles and femora of mice is determined. The results showed that LBP-4 is composed of six kinds of monosaccharides that can enhance food conversion rate and the content of zinc and iron in body of mice, and reduce the body weight.

Effects of polyamines on organogenesis and somatic embryogenesis of Lycium barbarum calli.

Levels of three endogenous free polyamines (PAs), Put, Spd and Spm, were detected during organogenesis and somatic embryogenesis (SE) of Lycium barabrum (L.) calli. The predominant forms of PAs in organogenesis and SE were found to be Put and Spd, respectively. In both developmental pathways, the changes of Put content were very similar, i.e., it accumulated quickly in the initial stages of calli differentiation and then decreased; with the further morphogenesis, increase in Put level was also observed. The highest level of Spd was obtained at day 1 of calli organogenesis; while, after 1 day of culture for calli SE, Spd level began to increase and reached a maximum at day 10. The treatments with exogenous PAs had positive effects on both organogenesis and SE, especially those of Spd on organogenesis and SE and Put on SE. However, Put showed no effects on adventitious bud formation. CHA, which inhibits the activity of Spd synthetase, prevented both adventitious bud formation and further development of somatic embryo into plantlet. Although MGBG, a specific inhibitor of S-adenosyl methionine decarboxylases, had little effects on organogenesis, it reduced the number of somatic embryo and the plantlets subsequently regenerated. Such inhibitions could be reversed by Spd (50 mumol/L). These results from organogenesis and SE of the same specie indicate that PAs influenced these two in vitro morphogenesis pathways.

We used callus of medical plant, Ningxia Lycium barbarum. L as material to induce somatic embryo genesis, and used multitracer technique and gamma-ray energy spectrum analytical method to study different concentration of AgNO3 on some absorption of trace metal ions during the somatic embryogenesis of Lycium barbarum. L. The results show that (1) when the concentration of AgNO3 is less then 50 mg/L, with the increase of AgNO3 concentration, the absorption rate of some metal ions is increasing, but when the concentration is excess of such concentration, the effects on the absorption rate are different. Ag+ has the effect of coordination, resistance or competition on the absorption of metal ions. (2) AgNO3 in suitable concentration will promote cell differentiation and somatic embryogenesis. When the concentration of AgNO3 is less then 50 mg/L, along with the increase of the concentration, the frequency of somatic embryogenesis is increased, and Ag+ has the promotion on the somatic embryogenesis of Lycium barbarum. L. While the concentration is up to 50 mg/L, it will mostly raise somatic embryogenesis in calli, the amount of callus is about 3 times as much as that of contrast group (in which AgNO3 is not added). When excess of this concentration point, Ag+ has toxic effect, and the somatic embryogenesis is inhibited remarkably.

The protein and DNA content in somatic embryos of Lycium barbarum L were analysed using quantitative stereologicai procedure formed from Cell Morphometry. The photos were transformed into digital images by the Sharp JX-610 scanner. Then the quantity of protein and DNA was count applying digital images processing software. By the result we discuss the relationship between changes in protein and DNA metabolize dynamic and somatic embryogensis.

A system of m (12.5% PEG8000): m (25% K2HPO4) = 1:1 was used to separate two kinds of standard proteins, glycoprotein in Morchella esculenta (L.) and glycoprotein in Lycium barbarum (L.), the upper phase was used as stationary phase, and the lower phase was used as mobile phase. The revolution speed was 500 r/min and the flow rate was 60 mL/h. In comparing with high performance liquid chromatography, cross-axis counter current chromatography can increase load capacity without loss of resolution. The advantage of this method was verified, indicating that cross-axis counter current chromatography was a useful method for biopolymer separation.

OBJECTIVE: To study the chemical constituents in the fruit of Lycium barbarum. METHOD: The chemical constituents were isolated by column chromatography and identified by spectral data. RESULT: The compounds obtained were identified as scopoletin(I), beta-sitosterol(II), p-coumaric acid(III), glucose(IV), daucosterol(V) and betaine(VI). CONCLUSION: Compounds III, IV and V were isolated from Lycium barbarum for the first time.

OBJECTIVE: To extract and isolate the active component inhibiting PC3 cell proliferation in vitro from the fruit of Lycium barbarum. METHOD: The active components of the fruit of L. barbarum were extracted by water, 95% ethanol, 50% ethanol respectively. Then, ethanol extract was extracted with acetone and n-butanol and isolated by SiO2 column and purified by TLC to obtain the active component. The chemical structure of active component was identified by the spectral analysis. MTT assay was used to compare the inhibition activity(IC50) of PC3 cells proliferation of the all extracts. RESULT: According IC50, the most active component among all extracts isolated from the fruit of L. barbarum isscopoletin. CONCLUSION: Scopoletin is the active component of the fruit of L. barbarum for inhibiting PC3 cell proliferation.

Hepatoprotective pyrrole derivatives of Lycium chinense fruits.

As a part of our search for hepatoprotective compounds from Lycium chinense fruits, three new pyrrole derivatives (1-3) were isolated. These compounds and a related synthetic methylated compound (4) were evaluated for their biological activity and structure-activity relationship, and compounds 1 and 2 showed hepatoprotective effects comparable to silybin at the concentration of 0.1 microM (64.4 and 65.8%, respectively).

[The protective effects of Lycium barbarum polysaccharide on alloxan-induced isolated islet cells damage in rats]

OBJECTIVE: To study the effects of Lycium barbarum polysaccharide (LBP) on alloxan-induced isolated islet cells damage in rats. METHODS: Isolated islet cells from SD rat were cultured in vitro. SOD activity and MDA content in rat islet cells incubated with alloxan or alloxan combined LBP were measured. RESULTS: Alloxan induced a significant decrease in SOD activity and a increase in MDA production, which were inhibited by LBP. CONCLUSION: LBP had protective effects on alloxan-induced isolated rat islet ceus damage.

Amino acid sequences of ferredoxins from Scopolia japonica and Lycium chinense: their similarities to that of Datura arborea.

The complete amino acid sequences of [2Fe-2S] ferredoxins from Scopolia japonica and Lycium chinense have been determined by automated Edman degradation of the entire Cm-proteins and of the peptides obtained by enzymatic digestions. These two ferredoxins exhibited only 2-7 differences in the amino acid sequence when compared to the Datura-ferredoxins (D. stramonium, D. metel, and D. arborea), and especially only 2 or 3 differences compared to D. arborea. On the contrary, 8-19 differences were observed among the other solanaceous ferredoxins. This suggests that S. japonica and L. chinense are closely related taxonomically to Datura plants, especially to D. arborea.

Protective effect of Fructus Lycii polysaccharides against time and hyperthermia-induced damage in cultured seminiferous epithelium.

Lycium barbarum L. (Solanaceae) is a Chinese medicinal plant whose fruits (Fructus Lycii) are used by Chinese physicians for treatment of infertility. However, the active ingredients and the mechanism of action underlying Lycium's fertility-facilitating effects remain unknown. Here we report that Fructus Lycii polysaccharides (FLPS) inhibit time- and hyperthermia-induced structural damage in murine seminiferous epithelium, in vitro. Moreover, we found that FLPS delayed apoptosis in this system, both at normothermic and hyperthermic culture conditions. Oxidative stress was reported to be a major cause of structural degradation and apoptosis in hyperthermic testes, and thus the protective effect of FLPS could implicate an antioxidant mechanism of action. To test this hypothesis we assayed the effect of FLPS on ultraviolet light-induced lipid peroxidation, and cytochrome c reduction by free radicals. We found that FLPS is a potent inhibitor of both of these reactions. Together, these results demonstrate the protective effect of FLPS on time- and hyperthermia-induced testicular degeneration in vitro, indicate the potential mechanism of action for this protective effect, and provide a scientific basis for the traditional use of this plant.

A new phenolic amide from Lycium chinense Miller.

A new phenolic amide, dihydro-N-caffeoyltyramine (1) was isolated from the root bark of Lycium chinense Miller, along with known compounds, trans-N-caffeoyltyramine (2), cis-N-caffeoyltyramine (3), and lyoniresinol 3alpha-O-beta-D-glucopyranoside (4). Their structures were determined by spectroscopic analysis. A NBT superoxide scavenging assay revealed that three phenolic amides showed potent antioxidative activity.

An ethnobiological field study on food plants and medicinal foods traditionally consumed in three Arbëresh (ethnic Albanian) communities in northern Lucania (southern Italy) document approximately 120 botanical taxa used for these purposes. Non-domesticated food vegetables (liakra), mostly gathered during the spring season, play a central role as traditional functional food. Quantitative ethnobotanical, ethnotaxonomical, ethnoecological, ethnogastronomical, and ethnopharmacological aspects related to gathering, processing, cooking and consumption of liakra are discussed. Unusual food species, such as Lycium europeaum, Centaurea calcitrapa, and a few spontaneous weedy Asteraceae and Brassicaceae species are locally used in the kitchen. Most of these are very poorly known phytochemically and phytopharmacologically. Moreover, an analysis of taste perception of the most commonly used botanical foods was conducted in the village of Ginestra. Arbëresh taste classification and indigenous criteria related to the perception of bitter taste in considering non-cultivated plants as food or medicine are discussed as well.

[A comparative study on the syntheses of DNA, RNA and protein during in vitro organogenesis and somatic embryogenesis of Lycium barbarum L.]

After the calli originating from the leaf explant of Lycium barbarum L. were selected and proliferated, the yellowish calli with same origin, similar state were transferred to O medium or E medium and the regenerative systems of organogenesis and somatic embryogenesis might form. By these systems, a comparative study on the synthetic activities of DNA, RNA and protein in the two in vitro regeneration pathways was carried out. The results were as follows: (1) Before meristemoid and embryogenic cells were formed, the synthesis of RNA was activated firstly, followed with the increase of synthesis rates of DNA and protein. During the formation of globular embryo, the synthesis rate of DNA increased quickly and then the activities of syntheses of RNA and protein reached the peak, while it was the contrary during germination of adventitious bud. (2) Components of soluble protein changed regularly. A peptide (153.6 kD) appeared during the initiation of both organogenesis and somatic embryogenesis. Several peptides disappearing gradually in the early-stage of differentiation could regenerate with the formation of shoot primordium and globular embryo. Corresponding to morphogenesis, both regenerative systems had specific peptides (84.9 kD, 46.3 kD and 44 kD, 36.2 kD) as molecular markers of its own development. In addition, the relation and mechanism of the two regenerative systems were discussed.

Three glycoconjugates, LbGp3, LbGp4 and LbGp5, were isolated from the fruit of Lycium barbarum L. Molecular weights of LbGp3, LbGp4 and LbGp5 were 9.25 x 10(4), 21.48 x 10(4) and 2.37 x 10(4), respectively. Carbohydrate contents of LbGp3, LbGP4 and LbGp5 were 93.6%, 85.6%, 8.6%, respectively. LbGp3 was composed of Ara and Gal in a molar ratio of 1:1. LbGp4 was composed of Ara, Gal, Rha and Glc in a molar ratio of 1.5:2.5:0.43:0.23. LbGp5 was composed of Rha, Ara, Xyl, Gal, Man and Glc in a molar ratio of 0.33:0.52:0.42:0.94:0.85:1. Elemental analysis of N contents: LbGp3 0.83%, LbGp4 1.72%, LbGp5 9.58%. The linkage between the glycan and protein may be of O-linkage in LbGp4.

Using Enzyme Linked Immunosorbent Assay (ELISA) method, we determined the ABA contents of different stages in somatic embryogenesis. The results showed that endogenous ABA contents increased to maximum value twice during somatic embryogenesis. After first maximum value of ABA contents embryogenic cells were observed in callus, and simultaneously, there was a specific protein of somatic embryogenesis investigated by SDS-PAGE. This protein accumulates preferentially in embryogenic callus but not in transferred callus. So it is suggested that ABA could promote the expression of specific genes and the synthesis of embryogenic protein during somatic embryogenesis in Lycium barbarum L. and ABA play an important role in globular stage as well. In addition, treatment of non-embryogenic activity callus with 4 mumol/L exogenous ABA could stimulate somatic embryogenesis. And the ABA function mechanism in relation to somatic embryogenesis was discussed.
